DOI : 10.5146/jcpath.2019.48 Multistep carcinogenesis is characterized by the occurrence of multiple cellular changes due to numerous genetic and non-genetic cellular changes resulting in clonal expansion and other features of malignancy such as invasion and/or metastasis. All cancers are considered to bear several characteristics that are also known as "hallmarks of cancer": self-sufficiency in proliferative signaling, insensitivity to growth suppressors, evasion of cell death, limitless replicative capacity, sustained/induced angiogenesis, invasion and metastasis capability, reprogrammed energy metabolism, evasion of immune response, constitution of a specific tumor microenvironment, genomic instability and mutation. In this review, mechanisms of carcinogenesis have been discussed focusing on basic and emerging hallmarks of cancer.
